After shooting I cock my rifle, remove the buddy bottle for filling and a few seconds after removing the bottle I hear a puff of air that  is coming from the regulator. Does this mean I have a blown o ring somewhere?

Not necessarily...
How does it shoot when you put the refilled bottle back on?
Leaking, velocity or POI changes??
Depending on how low you shot your bottle to..
It could be just air passing by the piston face seal!

The gun shoots fine.  I have checked and it also does this if I remove a newly filled bottle.

OK...
So if it dumps are when removing a full bottle...
a seal maybe involved....
KevG might need to jump in...
Hard to fully diagnose by Long-Distance..

Normal for mfr reg.some times they can hold air behind the reg with bottle removed for hours and some times can dump after a minute or two.if bottle as been shot lower than reg set pressure it will dump air as soon as you remove bottle as reg will be in open state.
